Abstract:
The dissertation presents the design of data acquisition system (DAQ) for fluorescence detection. The system is based on Silicon Photomultiplier (SiPM) with dedicated front-end ASIC. The emphasis has been placed on ensuring the best performance of the SiPM, especially a solution to stabilize the gain of the photodetector, which is temperature dependent. Implemented gain stabilization method enables to keep the gain constant with a σ below 1% within 10-40 °C temperature range. The designed DAQ with the SiPM has been used in fluorescence detection in stationary and microfluidic optic systems. Detection limits and calibration curves of two fluorophores (fluorescein and CF488) and antibody-fluorophor compound have been established. Influence of temperature on parameters of the designed application is strongly limited. The DAQ can immediately adapt to occurring temperature fluctuations keeping the gain of measured signal constant. This characteristic of the measurement system can be adopted in applications beyond the fluorescence domain e.g. in detection of high energy particles with scintillators. It is also an important element of a mobile application used in changing environmental conditions, especially in terms of temperature. The system has been successfully tested with SiPMs of various internal gain. The results presented in this dissertation have confirmed that designed measurement system has a potential use in biomedicine, especially in the field of biomarker detection.



Streszczenie:
W rozprawie doktorskiej przedstawiono projekt systemu akwizycji danych do badania fluorescencji. System oparty jest na krzemowym fotopowielaczu (Silicon Photomultiplier – SiPM) z dedykowanym układem scalonym ASIC. Zapewniono najlepsze warunki pracy fotodetektora, zwłaszcza w zakresie stabilizacji wzmocnienia, którego wartość jest zależna od temperatury. Wdrożona metoda stabilizacji umożliwia utrzymanie stałego wzmocnienia z σ poniżej 1% w zakresie temperatury 10-40 °C. Zaprojektowany system akwizycji danych (Data Acquisition System – DAQ) został wykorzystany do wykrywania fluorescencji w stacjonarnych i mikroprzepływowych układach optycznych. Ustalono granice wykrywalności i krzywe kalibracji dwóch barwników fluorescencyjnych (fluoresceiny i CF488) oraz związku przeciwciało-barwnik. Wpływ temperatury na parametry zaprojektowanej aplikacji jest silnie ograniczony. DAQ może natychmiast przystosowywać się do zmian temperatury, utrzymując stałą wartość wzmocnienia. Ta cecha systemu pomiarowego może znaleźć zastosowanie również w aplikacjach spoza badań fluorescencji, np. w wykrywaniu cząstek o wysokiej energii w systemach ze scyntylatorami. Jest to również istotny element aplikacji mobilnej stosowanej w zmieniających się warunkach środowiskowych, zwłaszcza pod względem temperatury. System został z powodzeniem przetestowany przy użyciu krzemowych fotopowielaczy o różnym wzmocnieniu nominalnym. Wyniki przedstawione w niniejszym rozprawie potwierdziły, że zaprojektowany system pomiarowy ma potencjalne zastosowanie w biomedycynie, szczególnie w zakresie detekcji biomarkerów.
